Purification of Aminoglycoside

چکیده

Sir: In 1967, UMEZAWA et al.1) found a kanamycin-neomycin phosphotransferase in Escherichia coli carrying R factor and proved that the resistance is directly related to the formation of this enzyme.2) In 1972, YAGISAWA et al.3) found another phosphotransferase in E. coli carrying a different R factor. These enzymes were different in their substrate specificities. The former which is now called aminoglycoside phosphotransferase I (APH(3')-I) transfers phosphate from ATP to the 3'hydroxyl group (the 3-hydroxyl group of the 6amino-6-deoxy-D-glucose moiety) of kanamycin and also to the 5"-hydroxyl group (the 5hydroxyl group of the D-ribose moiety) in lividomycin A but not to the 3'-hydroxyl

Purification and characterization of aminoglycoside-modifying enzymes from Staphylococcus aureus and Staphylococcus epidermidis.

Several strains of Staphylococcus aureus and Staphylococcus epidermidis, exhibiting characteristic resistance patterns to aminoglycoside antibiotics, were examined. The aminoglycoside-modifying enzymes from these strains were purified by DEAE-Sephadex A-50 chromatography, affinity chromatography, and Sephadex G-100 gel filtration. Three enzymes, a 3'-phosphotransferase III (molecular weight, 31...

Purification and characterization of aminoglycoside 3'-phosphotransferase type IIa and kinetic comparison with a new mutant enzyme.

Aminoglycoside 3'-phosphotransferase [APH(3')s] provide an important means for high-level resistance to neomycin- and kanamycin-type aminoglycoside antibiotics. A four-step purification which affords milligram quantities of homogeneous APH(3') type IIa [APH(3')-IIa] is described. The kinetic parameters for the turnover of five substrates by the enzyme were determined, and the pH dependence and ...

Investigation of high-level aminoglycoside resistance in Enterococcus spp. isolated from Caspian horse feces in Guilan province, northern Iran

Background and Aim: Due to the widespread use of antimicrobials in veterinary medicine and the increase in livestock production, it seems that the risk of spreading antibiotic resistance in human societies is more related to animals and the veterinary field. In this study, antibiotic resistance, high-level aminoglycoside resistance and frequency of aminoglycoside resistance associated genes amo...

Purification and structure elucidation of antifungal and antibacterial activities of newly isolated Streptomyces sp. strain US80.

A new actinomycete strain, designated US80 and producing antimicrobial activities against Gram-positive and Gram-negative bacteria and fungi, was isolated from Tunisian oasis soil. Cultural characteristic studies strongly suggested that this strain belongs to the genus Streptomyces. The nucleotide sequence of the 16S rRNA gene (1517 pb) of Streptomyces sp. strain US80 exhibited close similarity...

Prevalence of Genes Encoding Aminoglycoside Modifying Enzymes in Clinical Isolates of Klebsiella Pneumoniae in the Hospitals of Borujerd

Background and Aims: Given the importance of aminoglycoside resistance in nosocomial and community infections caused by bacterial pathogenes such as Klebsiella pneumoniae (K. pneumoniae), the aim of this study was to determine the frequency of aac (6')- Ib and aac (3)- IIa, the genes encoding aminoglycoside modifying enzymes involved in aminoglycoside resistance. Material and Methods: A t...
